Abstract	Cilj istraživanja bio je: (i) razviti matematičke modele električnih i toplinskih pojava u tkivu podvrgnutom djelovanju električnih impulsa kakvi se koriste za in vivo elektropermeabilizaciju i elektrogenski prijenos, i (ii) analizirati mogućnost da mjerenje napona i struje za vrijeme davanja impulsa može poslužiti za aktivno praćenje i predviđanje stupnja elektropermeabilizacije tkiva. Predloženi modeli temelje se na rješavanju odgovarajućih parcijalnih diferencijalnih jednadžbi za raspodjelu električnog potencijala i temperature u tkivu metodom konačnih elemenata u tri dimenzije. Za dvije konfiguracije elektroda (pločaste i par iglastih) provedene su simulacije za različite protokole davanja impulsa uključujući kratke visokonaponske permeabilizacijske impulse i duge niskonaponske elektroforetske impulse i vrednovani su utjecaji specifične električne vodljivosti tkiva (uključujući i anizotropnost, nelinearnost i temperaturnu ovisnost), parametara impulsa (amplituda, broj, trajanje, repeticija), perfuzije krvi i odvođena topline konvekcijom s površine prema okolini na raspodjelu električnog polja i temperature u tkivu te na rezultirajući valni oblik struje. Pored teorijskog rada provedeno je eksperimentalno ispitivanje predloženih modela mjerenjem struje i napona za vrijeme davanja permeabilizacijskih električnih impulsa i to in vitro na gustim suspenzijama stanica B16-F1 i in vivo na jetri štakora. Rezultati proračuna i predložene metode modeliranja i analize donose važne informacije primjenjive za određivanje parametara električnih impulsa i konfiguracija elektroda za elektrotransfekciju, s mogućom primjenom za učinkovitu i sigurnu nevirusnu gensku terapiju, kao i za poboljšanje sadašnjih protokola davanja impulsa za elektrokemoterapiju. Eksperimentalni rezultati ukazuju na mogućnost praćenja stvaranja pora u stvarnom vremenu i predviđanje ishoda elektropermeabilizacije tkiva temeljem informacije dobivene mjerenjem struje i napona za vrijeme davanja impulsa.
Abstract (english)	The aims of this study were: (i) to develop mathematical models of electric and related thermal phenomena in tissue exposed to electric pulses that are used for in vivo electropermeabilization and gene electrotransfer, and (ii) to analyze the possibility that voltage and current measurement during pulse delivery can be used to actively monitor and predict the extent of tissue electropermeabilization. The proposed models are based on the solution of appropriate partial differential equations for electric potential and temperature distribution in tissue using finite-element method (3D FEM). For two configurations of electrodes (parallel plates and two needles) and different pulse delivery protocols, including short high-voltage permeabilizing pulses and long low-voltage electrophoretic pulses, we performed numerous simulations in order to quantify the influence of tissue electrical conductivity (including anisotropy, nonlinearity and temperature dependence), pulse parameters (amplitude, number, duration and frequency), blood perfusion and free air convection from the surface, on the distribution of electric field and temperature in tissue and also on the resulting reaction current. In addition to theoretical work, we also performed experimental tests of our models by measuring current and voltage during delivery of permeabilizing electric pulses in vitro on dense suspensions of B16-F1 cells and in vivo on rat liver. Numerical results and proposed methods of modeling bring important information for the selection of pulse parameters and electrode configuration for DNA electrotransfer with promising application for efficient and safe non-viral gene therapy and also for the improvement of present pulse delivery protocols for electrochemotherapy. Experimental results indicate that voltage and current measurement during pulse delivery can potentially be used for real-time monitoring of pore formation and prediction of electropermeabilization outcome.
