Abstract
Polimorfizmi jednog nukleotida (SNP, engl. single nucleotide polymorphism) gena FOXO3 i FasL mogu biti povezani s razvojem i tijekom reumatoidnog artritisa (RA). SNP gena FasL povećava rizik oboljevanja od RA. Oba su gena uključena u apoptozu, pri čemu transkripcijski faktor FOXO3 supresijom Fas liganda sprječava apoptozu i podržava tijek kronične upale. Izolacijom DNA iz periferne krvi, određivanjem prisutnosti SNP-a u navedenim genima, određivanjem razine interleukina-10 (IL-10) i čimbenika tumorske nekroze α (TNFα, prema engl. tumor necrosis factor) iz krvi imunoenzimskom metodom ELISA, te korelacijom prisustva SNP-ova s promjenama razine citokina analizirani su učestalost polimorfizama, ekspresija proupalnih (TNFα) i protuupalnih (IL-10) citokina u bolesnika s reumatoidnim artritisom u odnosu na kontrolnu skupinu. Polimorfizmi gena FOXO3 i FasL u ispitivanom uzorku hrvatske populacije nisu promijenjene učestalosti u RA, te nisu jasno povezani su s promjenom ekspresije citokina IL-10 i TNFα.
Abstract (english)
FOXO3 and FasL single nucleotide polymorphisms (SNP) could be related to the course of rheumatoid arthritis (RA) due to changed expression of certain cytokines. FasL gene SNP increases the susceptibility to RA. Both genes are involved in apoptosis, with FOXO3 transcription factor suppression of Fas ligand preventing apoptosis and sustaining chronic inflammation. We determined the frequency of SNPs, expression of proinflammatory (TNFα) and anti-inflammatory (IL-10) cytokines in RA patients by methods of DNA isolation from peripheral blood, genotyping, determination of TNFα and IL-10 (ELISA) and than correlated polymorphisms with expression of cytokines. Results: In our cohort FOXO3 and FasL polymorphisms does not differ in frequency in RA, and are not related to expression changes of TNFα and IL-10 with consequences to the course of RA.
