Abstract	Cilj: Glavni cilj ovog istraživanja bio je imunohistokemijskim metodama iz uzoraka oralnih leukoplakija (OL) i eritroplakija (OE) te planocelularnih karcinoma usne šupljine (OSCC) odrediti stupanj izraženosti nEGFR-a i drugih markera staničnog ciklusa te njihov utjecaj na malignu transformaciju OL i OE kao i na kliničke ishode i prognozu bolesti oboljelih od OSCC-a. Materijali i metode: Imunohistokemijskim metodama analiziran je izražaj markera nEGFR, mEGFR, Ki-67, p53, ciklina D1 te ABCG2 u patohistološkim uzorcima 50 bolesnika s OL ili OE i 52 bolesnika s OSCC-om. Rezultati: Utvrđen je porast izražaja mEGFR i nEGFR razvojem bolesti (p<0,0001). U skupini bolesnika s OL i OE dokazali smo pozitivnu korelaciju između nEGFR-a i Ki67, p53, ciklina D1 i mEGFR, dok smo u skupini OSCC-a utvrdili pozitivnu korelaciju između nEGFR-a i Ki67, mEGFR (p<0,05). Istodobni izražaj nEGFR-a i ABCG2 povećava rizik od maligne transformacije 12,8 puta. Tumori bez perineuralne (PNI) invazije su pokazivali veću učestalost izražaja proteina p53 u odnosu na tumore s PNI (p=0,02), dok je za ABCG2 utvrđena pozitivna korelacija s istim parametrom (p=0,02). Bolesnici s OSCC-om i prekomjernim izražajem nEGFR-a imali kraće sveukupno doživljenje (p=0,004). Zaključak: Rezultati ovog istraživanja impliciraju na moguću važnu neovisnu ulogu nEGFR-a u oralnoj karcinogenezi.
Abstract (english)	Aim: The aim of this study was to determine the expression of nEGFR and other cell cycle markers and their influence on malignant transformation of oral leukoplakia (OL) and oral erythroplakia (OE) and on clinical outcomes and prognosis of patients with OSCC. Materials and methods: The expression of the markers nEGFR, mEGFR, Ki-67, p53, cyclin D1, and ABCG2 was analyzed by immunohistochemical methods in pathohistological specimens from 50 patients with OL or OE and 52 patients with OSCC. Results: An increase in the expression of mEGFR and nEGFR was found with the development of the disease (p<0.0001). In the group of patients with OL and OE we found a positive correlation between nEGFR and Ki67, p53, cyclin D1 and mEGFR, while in the group of patients with OSCC we found a positive correlation between nEGFR and Ki67, mEGFR (p<0.05). Co-expression of nEGFR and ABCG2 increased the risk of malignant transformation by 12.8-fold. Tumors without perineural (PNI) invasion had higher expression of p53 protein than tumors with PNI (p=0.02). Patients with OSCC and overexpression of nEGFR had shorter overall survival (p=0.004). Conclusion: The results of this study suggest a potentially important independent role of nEGFR in oral carcinogenesis.
